:
Introduction & Aim Since vitamin D is considered as an anticancer agent by controlling genes responsible for cells proliferation, differentiation, and apoptosis, biological and epidemiologic data suggest that vitamin D levels may influence breast cancer development. The vitamin D receptor (VDR) is a crucial mediator for the cellular effects of vitamin D and additionally interacts with other cell signaling pathways that influence cancer development. We intend to assess the relevance between VDR gene polymorphisms (Fok1 & Bsm1) with breast cancer in Saudi women. Material & Method Mutation analysis was done for two polymorphisms of the VDR gene; Fok1 & Bsm1 in a population of 50 Saudi women suffering from breast cancer and 50 control group. Result For Bsm1, 14/50 were positive for two mutations 11 in C\T and 3 in G\A. More than half of C\T mutation were found homozygote, 6 cases. For Fok1 17/50 were positive to A\G polymorphism and C-deletion at the same time 10 of them were heterozygote. One case shows a C-deletion mutation. Conclusion This study showed that there is a close relationship between the presence of mutations in this gene and the presence of breast cancer. This means that, mutations in this gene could be considered as diagnostic tool for breast cancer occurrence. Our results suggest that the. VDR may be a mediator of breast cancer risk and could represent a target for cancer prevention efforts.
